ABSTRACT

Unlike healthy, non-transformed cells, the proteostasis network of cancer cells is taxed to produce proteins involved in tumor development. Cancer cells have a higher dependency on molecular chaperones to maintain proteostasis. The chaperonin T-complex protein ring complex (TRiC) contains eight paralogous subunits (CCT1-8), and assists the folding of as many as 10% of cytosolic proteome. TRiC is essential for the progression of some cancers, but the roles of TRiC subunits in osteosarcoma remain to be explored. Here, we show that CCT4/TRiC is significantly correlated in human osteosarcoma, and plays a critical role in osteosarcoma cell survival. We identify a compound anticarin-β that can specifically bind to and inhibit CCT4. Anticarin-β shows higher selectivity in cancer cells than in normal cells. Mechanistically, anticarin-β potently impedes CCT4-mediated STAT3 maturation. Anticarin-β displays remarkable antitumor efficacy in orthotopic and patient-derived xenograft models of osteosarcoma. Collectively, our data uncover a key role of CCT4 in osteosarcoma, and propose a promising treatment strategy for osteosarcoma by disrupting CCT4 and proteostasis.

ABSTRACT

The neuroimaging provides a lot of useful information about the neuromechanism of restless legs syndrome (RLS).The voxel-based morphometry and diffusion tensor imaging findings reported brain microstructural abnormalities, which were more consistent concerning levels of white matter and belonged to the sensorimotor and limbic system.Functional magnetic resonance imaging (MRI) studies also demonstrated the neural activity and/or functional connectivity changes in the sensorimotor and limbic network.Positron emission tomography and single photon emission computed tomography studies supported the dysfunction of the dopaminergic pathways involving not only the nigrostriatal but also mesolimbic pathways.Iron-sensitive MRI verified low brain iron content mainly in substantia nigra and thalamus. The primary change might be the reduction of brain iron content, which leads to the dysfunction of the nigrostriatal and mesolimbic dopaminergic pathways and in turn to a dysregulation of sensorimotor and limbic network resulting in the symptoms of RLS.

ABSTRACT

Objective@#To investigate the changes in clinical characteristics and laboratory indexes before and after the termination of pregnancy in patients with acute fatty liver of pregnancy (AFLP).@*Methods@#Patients with acute fatty liver of pregnancy who had been admitted to the Department of Obstetrics and Gynecology at the Second Affiliated Hospital of Chongqing Medical University and Chongqing Municipal People’s Hospital of Jiangbei District between 2007 and 2018 were selected. Clinical characteristics and complications during diagnosis and treatment, changes in blood coagulation, liver and kidney function, and postpartum recovery were collected for retrospectively analysis.@*Results@#54 cases with average gestational age of 35.0±1.7 weeks at third trimester of pregnancy with AFLP were treated. The most common gastrointestinal symptoms were yellow urine, nausea and vomiting. All patients had elevated bilirubin. 90.7% patients had changes in blood coagulation function and 68.5% had elevated serum creatinine. Transaminase levels were dropped rapidly within 1-2 days after the termination of pregnancy. Total bilirubin recovery was slow and partially recovered after 6-8 days. Serum creatinine and BUN increased slightly after delivery, reaching a peak at 3-4 days and then began to deplete. There was slight change in prothrombin time and fibrinogen after delivery, but returned to normal level after 5-6 days. The most common complications were AKI (74.1%), LF (42.6%), PPH (40.7%) and DIC (33.3%). Twenty-three of the 54 cases (42.6%) progressed to acute liver failure. AFLP complicated with ALF course was significantly longer than healthy controls, and the disease severity was significantly increased, with a mortality rate of 17.4% (4/23), and 0 in healthy controls. The difference was statistically significant.@*Conclusion@#Early diagnosis and termination of pregnancy are the key factors to determine the prognosis of pregnant patients with acute fatty liver. Blood coagulation function does not deteriorate after termination of pregnancy and renal function begins to recover after 4 days with slight restoration of liver function. The control of complications is an important factor to determine the prognosis of patients.

ABSTRACT

OBJECTIVE: To compare inhibitory effects of ethanol extract of different medicinal parts (root, stem, leaf, seed, flower and flesh) from Syzygium jambos on the activities of α-glycosidase and α-amylase. METHODS: Using half-inhibitory concentration value (IC50) as evaluation index, acarbose as positive control, inhibitory effects of ethanol extract of different medicinal parts from S. jambos on the activities of α-glycosidase (from yeast and small instestine in mice) and α-amylase were evaluated with in vitro inhibition model. The enzymatic dynamics and Lineweaver-Burk methods were used to analyze the inhibitory type of the best medicinal part on the activities of α-glycosidase and α-amylase. RESULTS: In the yeast α-glucosidase inhibitory activity test, the order of inhibitory activity was S. jambos seed>S. jambos stem>S. jambos leaf>S. jambos root>S. jambos flower>S. jambos flesh>acarbose. In the mice intestine α-glucosidase inhibitory activity test, the order of inhibitory activity was S. jambos seed>S. jambos stem>S. jambos root>S. jambos leaf>S. jambos flower>S. jambos flesh>acarbose. In the α-amylase inhibitory activity test, the order of inhibitory activity was acarbose>S. jambos seed>S. jambos stem>S. jambos root>S. jambos leaf>S. jambos flesh>S. jambos flower. Ethanol extract of S. jambos seed had the stronger inhibition activity against α-glucosidase from yeast,α-glucosidase from small intestine in mice and α-amylase than other medicinal parts [IC50 were(6.64±0.24), (32.77±2.46) and (41.18±1.63) μg/mL]. Ethanol extract of S. jambos seed had the stronger inhibition activity against α-glucosidase than acarbose [IC50 to α-glucosidase from yeast and α-glucosidase from small intestine in mice were (2 833.33±5.48), (1 304.21±6.45) μg/mL] (P<0.05). The inhibitory effect of ethanol extract from S. jambos on the activity of α-amylase was less than that of acarbose [IC50 was (27.27±1.24) μg/mL] (P<0.05). Enzymatic dynamics showed that the inhibitory type of ethanol extract from S. jambos seed on α-glucosidase and α-amylase were both reversible competitive inhibition. CONCLUSIONS: Among different parts of S. jambos such as root, stem, leaf, seed, flower and flesh, S. jambos seed shows the strongest inhibitory effects on the activities of α-glucosidase and α-amylase, which has the value of being developed for the treatment of diabetes or health food.

ABSTRACT

OBJECTIVE: To investigate in vitro antibacterial activity of the extracts from the root, stem and leaves of Psychotria rubra, and to investigate its mechanism of antibacterial effects. METHODS: 95% ethanol extracts from the root, stem and leaves of P. rubra were used to prepare solution with mass concentration of 100 mg/mL (calculated by extract). Using as penicillin sodium (0.5 mg/mL) and streptomycin sulfate (128 mg/mL) as positive control, plat stiletto method was used to determine antibacterial effects of the extracts from different parts of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Micrococcus luteus and Enterococcus faecalis. Using systematic solvent extraction method, the petroleum ether, ethyl acetate and n-butyl alcohol were used to extract antibacterial activity parts from P. rubra as ad to obtain the extracts of corresponding polar parts. After preparing 100 mg/mL drug solution (calculated by extract), antibacterial effects of above 6 kinds of bacteria were investigated. The micro-broth dilution method and agar culture medium plate method were used to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC), and screen polar parts with bacteriostasis and drug-sensitive strains. Under the concentration of 0.5MIC and MIC, the growth curves of sensitive bacteria were drawn (treated for 36 h, every 4 h); the contents of alkaline phosphatase (AKP) and soluble protein were detected in suspension (treated for 10 h, every 2 h); bouillon culture medium instead of the extract as blank control. RESULTS: 100 mg/mL ethanol extracts from the root, stem and leaves of P. rubra showed good antibacterial effect to above 6 kinds of bacteria, in descending order as stem>leaves>root. Among different polar parts from the stem of P. rubra, antibacterial effect of the ethyl acetate extract was best, especially for the S. aureus, the diameter of inhibition zone reached (38.93±0.12) mm, and both MIC and MBC were 0.39 mg/mL. The ethyl acetate extract could significantly inhibit the proliferation of S. aureus, and its alkaline phosphatase and protein content in suspension were increased significantly compared with blank control (P<0.05). CONCLUSIONS: The stem from P. rubra is effective antibacterial parts, and exhibit good antibacterial activity to 6 kinds of common pathogens. The ethyl acetate extract from the stem of P. rubra shows strongest antibacterial effect on S. aureus, and could gradually destroy the integrity of cell wall and cell membrane.

ABSTRACT

This article was aimed to study the protection effects of Pa-Bing Formula No. 2 (PBFN-2) on neurons of substantia nigra in Parkinson's disease (PD) rats models in order to explore the possible mechanism of PBFN-2 in PD treatment. Rats were stereotaxically injected with 6-hydroxydopamine (6-OHDA) solution into the left stria-tum in two-site. Rat showed consistent right whirling and the number of rotation was more than 7 r·min-1 induced by APO in 30 min, then the rat was judged as PD model. A total of 14 rats modeled successfully were randomly di-vided into the model group and the treatment group. At the same time, the normal group and sham operation group were also established. Same volume of distilled water was given to the normal group, sham operation group and the model group. PBFN-2 (32.0 g·kg-1) was given to the treatment group for 4 weeks. Microscope was used to observe pathological changes of substantia nigra by nissl stain and changes of TH, Nrf2 and HO-1 immunohistochemical stain. The results showed that compared with the normal group, the nissl bodies were badly injured. Expressions of TH-positive cells were obviously reduced (P<0.01). The expression of Nrf2 nucleus protein and HO-1 protein were obviously increased in substantia nigra of PD rats in model group (P<0.01). Compared to the model group, PBFN-2 effectively increased nissl bodies in neuronal cells of substantia nigra of PD rats, and elevated the number of TH-immunoreactive cells in substantia nigra (P<0.05). The expressions of Nrf2 nucleus protein, HO-1 protein were significantly up-regulated (P<0.05). It was concluded that PBFN-2 had an obvious neuroprotection on the neuronal cells in substantia nigra of PD rats induced by 6-OHDA. The underlying mechanisms may be associated with regu-lation of Nrf2 nucleus protein and HO-1 protein expressions.

ABSTRACT

Parkinson's disease (PD) is a commonly encountered central neurodegenerative disease in elderly people. According to theories of traditional Chinese medicine (TCM), PD is characterized by deficiency in the root and excess in the branch. Deficiency was referred to qi-blood deficiency of the liver and kidney. Excess was referred as the wind, fire, phlegm and stasis. Deficiency was the root and excess was the branch. Good efficacies have been obtained by treatment based on syndrome differentiation, treatment with specific prescriptions, acupuncture and moxibustion and comprehensive treatment. Some experiments had been conducted to elucidate its mechanisms. However, no uni-fied standard for therapeutic evaluation, poor control of medicinal quality, inferior quality of designed clinical trials, and unclear treatment mechanism in the clinical study on therapeutic effect of PD treatment with TCM require fur-ther studied.

ABSTRACT

An analytical method for the simultaneous determination of four quinolones in soil was developed. Soil samples were extracted by a mixture of 50% magnesium nitrate and 10% ammonia(96∶ 4, V/V)with an ultrasonic-assisted extraction, then purified and concentrated by HLB cartridge, and eluted with acetonitrile and 0.067 mol/L phosphoric acid(5∶ 1, V/V). Using acetonitrile and 0.067 mol/L phosphoric acid(pH=2.5) as the mobile phase, these analytes were quantificated by HPLC(fluorimetric detector) at excitation and emission wavelength of 280 nm and 450 nm respectively. The detective limits for four quinolones in soil were from 0.58 to 0.03 g/kg. The recoveries were 60.4% to 99.3% for soil samples. The method was successfully applied to determine the quinolones in soil samples from vegetable fields. Four quinolone compounds were detected to a different extent with total amounts of quinolones ranged from 27.84 to 129.26 μg/kg.

ABSTRACT

This was a study aimed to observe the proliferating ability inhibited by energy controllable steep pulse (ECSP) and to detect the expression of gene with relation to the proliferating ability of the tumor in breast cancer cell line; the possible mechanisms were also addressed. Human breast cancer cell line MDA-MB-231 was treated with ECSP; the apoptosis and the expression of tumor suppressor gene--Rb genes and E2F1 genes in ECSP group and control group were detected by TUNEL staining and Reverse Transcripitional PCR respectively. ECSP was found to inhibit the proliferating ability of breast cancer cells markedly, the cell amount in ECSP group decreased and the TUNEL positive cells increased obviously, compared to control; 24 hours after treatment the expression of Rb genes mRNA increased, whereas the expression of E2F1 mRNA decreased. These findings indicate that the proliferating ability of breast cancer cells can be inhibited by ECSP markedly, the apoptosis of breast cancer cell can be induced by ECSP, and the Rb genes and E2F1 genes may be involved in the course.
